چکیده
Abstract Background Epileptogenesis can be defined as the process by which a previously healthy brain develops tendency toward recurrent electrical activity, occurring in three phases: first an initial trigger (such stroke, infections, and traumatic injury); followed latency period onset of spontaneous seizures characterizes epilepsy. Main body The mechanisms that may involved epileptogenesis are inflammation, neurogenesis, migration neurons to different regions brain, neural reorganization, neuroplasticity. In recent years, experimental studies have enabled discovery several epileptogenesis, mainly neuroinflammation, involves activation glial cells increase specific inflammatory mediators. lack animal model protocol for epileptogenic compounds contributes difficulty understanding disease development creation new drugs. Conclusion To solve these difficulties, approach is needed AEDs focus on consolidation models antiepileptogenic compounds, aiming reach clinical phases study. Some examples rapamycin, inhibits mTOR signaling, losartan, potentiates effect some AEDs. Based this, this review discusses main well its pharmacological approach.

Immune mechanisms in epileptogenesis
Epilepsy is a chronic brain disorder that affects 1% of the human population worldwide. Immune responses are implicated in seizure induction and the development of epilepsy. Pre-clinical and clinical evidence have accumulated to suggest a positive feedback cycle between brain inflammation and epileptogenesis. Prolonged or recurrent seizures and brain injuries lead to upregulation of proinflamma...
متن کاملNeuronal mechanisms of epileptogenesis
The primary purpose of this topic is to collect scientific contributions providing novel insights in the cellular and molecular mechanisms of epileptogenesis as potential targets for innovative therapeutic approaches aimed at preventing the chronic epileptic disorder. Prevention of chronic epileptic disorder with an appropriate intervention might represent the most ambitious goal in the clinica...
متن کامل'Non-synaptic' mechanisms in seizures and epileptogenesis.
The role of 'non-synaptic' mechanisms (i.e. those mechanisms that are independent of active chemical synpases) in the synchronization of neuronal activity during seizures and their possible contribution to chronic epileptogenesis are summarized. These 'non-synaptic' mechanisms include electrotonic coupling through gap junctions, electrical field effects (i.e. ephaptic transmission), and ionic i...
متن کاملGenes and molecular mechanisms involved in the epileptogenesis of idiopathic absence epilepsies
Idiopathic absence epilepsies (IAE), that have high prevalence particularly among children and adolescents, are complex disorders mainly caused by genetic factors. Childhood absence epilepsy and juvenile absence epilepsy are among the most common subtypes of IAEs. While the role of ion channels has been the primary focus of epilepsy research, the analysis of mutation and association in both pat...
